Sažetak
Decidual lymphocytes (DL) are non-adherent cell fraction obtained by trypsin digestion of normal human first trimester pregnancy decidua containing mostly non-classical CD3-CD16-CD56++ NK cells full of perforin. There is very few data about the basic mechanisms regulating perforin expression and cytotoxicity of DL, although the significance of local events is emphasised, including the role of decidual macrophages. The aim of this study was to investigate the phenotype of decidual adherent cells (DAC), also obtained by trypsin digestion of decidual tissue and possible effects of these cells in regulation of perforin expression and cytolytic activity of DL. Immunohistochemistry staining of DAC showed that about 80% of DAC are vimentin+,  actin+, Epidermal Growth Factor Receptor+ but cytokeratin- stromal cells and about 10-15% are Ber-Mac-3+ macrophages. By double staining flow cytometric analyses we found that decidual macrophages are also HLA-DR+ (activated). DAC suspension is responsible for upregulation of perforin expression in DL during 18hrs cultivation in vitro. However, HLA-DR depleted DAC is loosing this potential, showing that decidual macrophages are cells responsible for the observed effect. Decrease of number of P+ cells in the suspension of DL following 18 hrs cultivation in vitro was accompanied by decreasing of cytolytic activity of DL measured by 2 hr PKH26 (red) cytotoxicity assay against K562 cell line. Further decrease in cytolytic activity was obtained in DL cultured 18 hrs in the supernatant of DAC. These results suggest that macrophages in vitro are responsible for upregulation of perforin expression, and at least partially for suppression of cytolytic activity of DL.
