Sažetak
The appearance and accumulation of perforin positive (P+) decidual large granular lymphocytes, mostly NK like cells (CD3-CD16-CD56++) in the first trimester normal human pregnancy decidua is progesterone dependent. Progesterone could be also one of the factors controlling cytolytic armentarium and cytotoxic potential of decidual lymphocytes (DL). These effects of progesterone could be indirect and mediated either by induction and upregulation of cytokines of Th2 type and/or by PIBF, a 34 kD protein secreted by progesterone receptor (PR) positive cells upon ligation with progesterone. In decidua PR+ cells are stromal cells. The aim of our study was to investigate whether progesterone and PIBF affect perforin expression and cytolytic activity of DL. Progesterone when added in the coculture of DL and decidual adherent cells - DAC (mostly stromal cells and macrophages) blocks upregulation of perforin expression mediated by DAC. DL from the suspension of decidual cells cultivated overnight with increasing concentrations of progesterone showed lower cytotoxicity against NK sensitive line K562, in a 2 hr PKH-26 (red) cytotoxicity assay. The number of P+ cells and the content of P per cell did not change significantly during cytotoxicity test, indicating that the block in the granule exocytosis occurred. The same results in vitro we obtained by PIBF. We also found by immunohistology many PIBF+ cells in the first trimester normal human pregnancy decidua. These results suggest that PIBF, as mediator of progesterone actions operates at maternal-fetal interface and could influence perforin expression and cytolytic activity of decidual lymphocytes in vivo.
