In early pregnancy decidua CD3-, CD56bright+, CD16- large granular lymphocytes (dLGL) are far the largest population of decidual lymphocytes. dLGL in their granules contain high levels of cytoloytic protein perforin (Pbright). Decidual NK activity was found to be confined to this particular cell type. It is a general opinion that dLGL have an important role in immune surveillance by clearing cells infected by viruses or others intracellular parasites, transformed or neoplastic cells and restraining growth of trophoblastic cells. Direct cytotoxic activity could be a mechanism involved. The aim of this study was to investigate the role of Th1 cytokines in the control of natural killer (NK) activity of decidual lymphocytes (DL). Cytolytic activity of unstimulated and cytokine stimulated (IL-2, IL-12 and IL-15) DL, obtained by elective termination of pregnancy was tested against K562 cell line in 2hr cytotoxicity PKH26 (red) test. The frequency of P positive cells and P content/cell were measured simultaneously. We have shown that DL have cytotoxic activity against classical NK cell targets and IL-2 and IL-12 can augment it. In contrary IL-15 can not increase cytotoxic activity of DL. These results suggest obvious difference between Th1 type of cytokines in the regulation of decidual cytotoxic potential.
