Sažetak
Cytokines of Th1 family, such as IFNgama, IL-2, IL-12, TNFalfa are considered detrimental to the maintenance of pregnancy, whereas the Th2 cytokines (IL-4, IL-6, IL-10, IL-13) have been suggested to be beneficial. Decidual large granular lymphocytes (dLGL), which contain high level of cytolytic protein perforin (P), serve an immune surveillance function protecting from microbial attack, malignant transformation and cytotrophoblast invasion. Our previous results showed that the interactions between dLGL and decidual macrophages (dM) play an important role in regulation of perforin: perforin can be upregulated by the addition either dM or their supernatants. The aim of our study was to investigate whether this regulation of P expression by dM is mediated by cytokines IL-10, IL-12 and IL-15. Antibodies anti IL-10, but not anti IL-12, were efficient in preventing the effects of dM on maintaining the P level in DL. Surprisingly, effects of dM and their supernatants on P expression in DL can be blocked completely by the addition of monoclonal anti IL-15 antibody. Contrary, P expression in PBL can not be blocked by the addition of anti IL-15 antibody. The results are pointing to the importance of macrophages and IL-15 in the control of activation of perforin mediated cytolytic pathway in decidua.
