Sažetak
Cattle genotype significantly influences meat and milk fatty acid composition and content. Consumers especially highlight food related health implications. Using molecular methods of specific gen allele detection it is possible to get more completed insight in allele population frequencies and effect on production. On BTA14 centromeric region is detected DGAT1 gene encoding diacylglycerol 0-acyltransferase, a microsomal enzyme that catalyses the final step of triglyceride synthesis. Earlier researches reported DGAT1 gene as candidate gene important in cattle marker assisted selection. The aim of this research was to determine allele frequencies of DGAT1 gene in different cattle populations in Croatia. The research included six cattle breeds. From randomly chosen individuals were collected tissue samples, DNA isolation was followed by PCR sequencing, PCR-RFLP restriction and gene polymorphism detection. The highest frequency of DGAT1 A allele was found in Hereford and Simmental population (0, 74 ; 0, 73), while significantly lower frequency was found in Holstein and Brown breed (0, 22 ; 0, 35). Istrian cattle and Charolais breeds showed more even A and K allele frequencies (0, 64 ; 0, 57). Results of the research gave basic for further analysis of DGAT1 gene effect on production performance and can be used in breeding programs.
