Sažetak
The aim of the study was to assess changes in the concentrations of fructosamine and malondialdehyde in diabetic NOD mice treated with acarbose and plant mixture extract P-9801091. The possible sources of oxidative stress and damage to protein in diabetes include free radicals generated by auto-oxidation reactions of sugars and sugar adducts to protein, and by auto-oxidation of unsaturated lipids in plasma and membrane proteins. Diabetes mellitus was induced in NOD mice by i.v. injection of alloxan, in a dose of 65 mg/kg, 7 days before the beginning of treatment. The treatment with acarbose (25 mg/100 g chow) and plant mixture extract P-9801091 (20 mg/kg b.w.) admixed to chow took 7 days. Acarbose is a complex oligosaccharide which reversibly inhibits ƒŃ-glucosidases present in the brush-border of the small intestinal mucosa, that are responsible for the digestion of complex polysaccharides and sucrose. The plant mixture extract P-9801091 (Zagreb, Croatia) was obtained by ethanol extraction followed by alcohol evaporation and lyophilization from the following plant drugs: Centaurii herba, Cichorii radix, Juniperi fructus, Millefolii herba, Myrtilli folium, Phaseoli pericarpium, Taraxaci radix, Urticae herba, Valerianae radix, and Murii folium. Serum level of fructosamine was lower in diabetic NOD mice treated with either acarbose or plant mixture extract P-9801091 than in diabetic NOD mice (373.5*62.0 *mol/L vs. 471.8*106.7 *mol/L for acarbose ; and 240.2*73.6 *mol/L vs. 347.4*26.3 *mol/L for plant mixture extract P-9801091). Malondialdehyde, a stable end-product of lipid peroxidation, was measured in liver homogenate, and was also found to be lower in diabetic NOD mice treated with either acarbose or plant mixture extract P-9801091 than in diabetic NOD mice (7.8*1.3 *mol/g vs. 9.8*5.8 *mol/g for acarbose ; and 13.2*5.7 *mol/g vs.22.2*9.9 *mol/g for plant mixture extract P-9801091). We believe that the inhibition of lipid peroxidation in diabetic NOD mice was caused by the hypoglycemic effect of acarbose or plant mixture extract P-9801091.
