Sažetak
Histochemical and immunohistological analysis of the dental pulp and periapical tissue in situ on the larger bones of mammals, such as the dog, is very difficult to perform on decalcified specimens. In our experiment we used method, known in osteology, for preparation of undecalcified bones. We induced pulpitis and periapical periodontitis on dogs premolar teeth by pulp exposure and left pulp chambers open for 50 days. As enamel is the greatest problem when preparing undecalcified teeth, the examined teeth were in situ decoronated. After dehydration the tissue was perfused with Xylene for 72 hours and then gradually perfused with a mixed solution of methylmethacrylate (MMA), dibutyl phthalate and benzoyl peroxide. For sectioning the blocks (5-7 ěm thick) we used a rotating microtome for bone Leica RM 2155, equipped with a tungsten carbide knife. Apart from the basic histological staining (toluidine blue) it was also possible to carry out histochemical and immunohistochemical analyses by this method, which cannot be performed on larger bones, demineralized with stronger acids such as formic, nitric or trichloracetic. This method of preparation is suitable for detailed quantitative and qualitative cytological analysis of changes in the pulp and periapex in situ with a light microscope. Several types of staining can be applied on the same tooth, which is difficult to achieve with the grinding method. Analysis of specimens showed exudative apical periodontitis on most teeth, although pulp vitality was maintained after experimental period of 50 days.
