Sažetak
The aim of study was to discriminate and compare the retention of non-fibrous inert particles and toxic fibers in the rat lung compartments during the subchronic exposure phase as contribution to elucidation of mechanisms underlying the lung clearance. The parameter we sought on the first, the seventh and the thirtieth postinsillation day was the quantity of particulate burdens inside the alveolar space, lung interstitium and lymphonodal tissue of polystyrene particles and the comixture of particles and asbestos fibers. After the lung lavage, lymphonodes were separated from the lung tissue. The obtained biological material was chemicaly dissolved. The particulate burden from lung compartments was separated on Nucleopor filters, and the forty-five fields of vision were randomly examined microscopically. The average number of particles per field was calculated for the filter samples. The study results revealed that the majority of particles was found in the alveolar compartment on day seven in the control animal group which received inert particles only. The highest lung tissue burden of particles was noted on day thirty, and the most effective accumulation of particles in the lymphonodal tissue was obtained on day seven in the same animal group. In comparison, the lung tissue burden of particles in the group of animals which received the comixture was the highest on the first postinstillation day, and remained high throught entire experiment. The same animal group showed a slow increasing tendency of particle accumulation in the lymph nodes until the last experimental day. It could be concluded that even in the subchronic phase of exposure, asbestos fibers might cause longer retention of material in the lung tissue, and their decreased accumulation in regional lymphnodes.
