Sažetak
The capacity of p53 protein to detect and appropriately respond to many cancer-related stresses is essential for prevention of malignancies in humans. Although it is not known how such diversity of stress signals can be integrated by a single molecule, common features of some p53-inducing stresses is compromised ribosome biogenesis and nucleolar structure. A few ribosomal proteins (RPS) have been suggested as transducers of p53-activating signals in response to pharmacologic inhibition of ribosome biogenesis. Here we report that a number DNA damaging stresses, including cytostatic drugs, also utilize RPL5 and RPL11 to activate the p53 tumor suppressor.Thus, mutations in these RPS might lead to malignant transformation, tumor progression and resistance to anti-cancer therapies.
