Sažetak
In the process of pre-mRNA splicing in mammals, exon recognition by spliceosome is assisted by members of the serine-arginine (SR) family of proteins, which specifically bind certain short regions in exons, called exon splicing enhancers (ESEs). Loss of an exonic regulator can lead to skipping of that exon, formation of a truncated protein and development of genetic diseases. Short length, degeneracy and context-dependence of these regulators make the biological evaluation of ESEs predicted by existing in silico tools difficult without added functionality of enriching query sequence with more data. We created the ESE Analyzer Web Server for improved evaluation of predicted ESEs based on the alignments of human paralogous sequences, observation of splicing regulators’ conservation patterns and integration of exon-specific data from different sources. It explores evolutionary selection on the level of exonic splicing regulators (coding DNA/RNA sequence) compared to selection on the level of protein sequence, using protein domains as model set of data. Potential sites of the conflicts between two selection pressures might be of evolutionary interest. Data are displayed in visually clear and comprehensive manner.
