Sažetak
Aquaculture of tuna is the fast growing world aquaculture sector. It constitutes a significant part of Croatian aquaculture and plays an important role in export trade. Despite economic importance, little is known about research of tuna diseases. In fish health management the most useful approach is prevention of disease and monitoring of the microorganisms. In contrast to many reports on the microbiology of other fish species there is a shortage of information available for cultured tuna particularly for bluefin tuna, Thunnus thynnus. Generally, main data described parasitofauna and parasitic diseases, while the data on bacterial and viral diseases are sporadic. Available data describe presence of bacteria only in the context of pathological conditions and disease outbreaks in tuna. Appearance of disease in humans as a result of poisoning by eating unsuitable tuna meat is better described, especially with histamine producing bacteria. The aim of this work was to investigate the initial microbiota of bluefin tuna in the Adriatic Sea. Ten bluefin tunas weighing 6.5 to 20.5 kg were obtained from tuna farm in the Middle Adriatic. Tuna samples were taken in spring and autumn from 2003 to 2008. Samples for bacterial isolation were collected from skin and gills swabs using sterile cotton stick (Eurotubo, Spain), as well as from spleen, liver and kidney using sterile bacterial loop. All samples were cultured on Tryptic soy agar (BBL) supplemented with 1 % NaCl. Isolated strains were further characterized using the API kits (bio Mérieux, France) and identified by the APILAB Plus identification software (bio Mérieux, France).Gram-negative bacteria dominated the bacterial composition of tuna. The bacterial load was higher in skin and gill samples than in the internal organs. One of the reasons may be the water quality, which was close to optimum and bacterial flora on tuna reflects the conditions in aquatic environment. The genera Brevundimonas, Klebsiella, Moraxella, Pasteurella, Pseudomonas, Staphylococcus, Vibrio and Weeksella present the initial microflora on examined tuna. Detection of Klebsiella pneumoniae in initial microflora of tuna is possible threat for histamine fish poisoning in humans.
