Sažetak
The sea bass (Dicentrarchus labrax) is one of the major commercial species in Croatian mariculture. Sea bass farms dominate along the eastern coast of the Adriatic Sea. Diseases outbreaks, as well as number and type of bacterial pathogens have been well described for some fish species, while the available literature on the bacterial flora of sea bass are scarce. The meat quality makes sea bass popular and highly valuable throughout the world. On European market demand for fresh sea bass in the last decade is increased and thus the need for the research of bacterial community. The objective of this study was to understand the microbial community associated with the culture system of sea bass, Dicentrarchus labrax and to determine the presence of potentially pathogenic bacteria in rearing systems along Croatian coast. Sampling was conducted on three sea bass farms in the area of southern, central and northern Adriatic. Fish samples were taken in spring and autumn in period of 2003 to 2008 from 360 randomly sampled fish. Samples for bacterial analyses were collected from skin and gills swabs using sterile cotton stick (Eurotubo, Spain), as well as from spleen, liver and kidney using sterile bacterial loop. All samples were cultured on Tryptic soy agar (BBL) supplemented with 1 % NaCl. Isolated strains were further characterized using the API kits (bio Mérieux, France) and identified by the APILAB Plus identification software (bio Mérieux, France). The bacterial genera were predominantly Gram-negative and comprised of Moraxella, Pasteurella, Nefermentor and Ochrobacter, while Staphylococcus, Micrococcus and Corynebacterium were the dominant Gram-positive bacteria. Also, during this research Vibrio alginolyticus was isolated, molecularly identified and confirmed by sequencing at three fish farms. Determined bacterial community associated with sea bass culture systems is important because the host–microbe interactions have implications on fish health, development and outbreaks of disease. Some of the isolated and identified bacteria may be opportunistic pathogens causing diseases in stressed fish populations, especially during larval development.
