Sažetak
Aims: The aim of this study was to detect the presence of periodontopathogens in the subgingival microbiota in patients with two different brackets type. (self-ligating and conventional brackets). Materials and methods: Thirty patients aged between 12 and 17 years participated in this study. 20 subjects with self-ligated (Damon 3MX, Ormco Corp., Glendora, , CA, USA), and 10 with conventional (bracket, Sprint, Forestadent, Pforzheim, Germany) brackets for at least 3 months. Subgingival plaque samples were collected in dry field conditions with sterile paper points for 20s and immediately transferred from the periodontal sulcus into a transporting box from the proximal sites of the 5 index teeth. These 5 samples were pooled so that the results were species specific and not site specific. The plaque samples were sent to a specialized laboratory using micro-Dent®test (Hain Lifescience GmbH, Nehren) which combine DNA amplification with subsequent detection using DNA probes. Once the bacterial DNA has been isolated bacterial detection was performed by polymerase chain reaction. The presence of five putative periodontopathoagenic species were detected: Actinobacillus actinomycetemcomitans, Porphyromonas gingivalis, Prevotella intermedia, Bacteroides forsythus and Treponema denticola. The individual pathogens were marked as: (0) undetected, (+) slightly positive, (++) positive, (+++) strongly positive. Statistical analysis was performed using descriptive and nonparametric analysis using the Mann Whitney test. Results: According to Mann Whitney test there was significant difference between Damon3MX and Sprint brackets (p=0.0006) for. A. actinomycetemcomitans, while other species did not show significance between the groups. 80% subjects with conventional brackets had positive or strongly positive count of A. actinomycetemcomitans , while in Damon group the result was only 15%. Conclusion: There was significant difference between Damon and conventional brackets regarding the presence of A. actinomycetemcomitans . Other species showed higher, but not significant values in subject with conventional brackets
