Sažetak
Human papillomavirus type 16 (HPV16) is the most common type found in cervical cancer. Variation of HPV16 has been extensively investigated except within the E1 region. We have investigated in more detail a novel HPV16 variant with 63nt duplication at position 1374 of the genome within the E1 region. A total of 822 HPV16 cervical samples, were analyzed for E6 variability using high resolution melting (HRM) and DNA sequencing, while the E1-1374^63nt duplication was assayed by specific PCR. The E1-1374^63nt duplication was found in 77 of 741 (10.4%) samples. The E-G350 was the most common variant (51.8%) followed by the reference variant (31.3%) among the 758 successfully analyzed samples. The whole genome sequence revealed 16 mutations specific to this variant with 4 mutations within the E1 region being unique to this variant and observed for the first time. The E1-1374^63nt duplication was exclusively found together with 350G mutation, with or without other E6 mutations. The most common E1-1374^63nt duplication variant was 350G monovariant (6.6% ; 49/741). This variant was the third most common variant overall and it was significantly associated with low-grade cervical lesions (p=0.028) when compared to the referent variant. Other E1-1374^63nt variants were found in <3.1% of samples, while other variants without the duplication were found in <1.3% of samples. Furthermore, in comparison with the prevalence of the most common types in Croatia (HPV 6/11, 16, 18, 31, 33, 45, 52, 58), this E1 variant was found more often than HPV45, which is the 3rd most common type found in cervical cancer worldwide. It could be assumed that the E1 variant is also more prevalent than other less common HPV types. Our findings show exclusive linkage of the E1-1374^63nt duplication and the 350G mutation. Thus, this variant is potentially clinically relevant considering its unusually high prevalence.
