Sažetak
The aim of this paper was to identify OTA-DNA adduct in blood and/or kidney of Serbian, Croatian, Bulgarian individuals exposed to OTA via food. OTA metabolites in blood and urine were analysed. DNA adduct were analysed by classical postlabeling technique and using the new high sensitive method allowing us to identified by LC/ms/ms the C8-2’-deoxyguanosine OTA (1 ; 2). In parallel, kidney DNA adduct and OTA metabolites in fluids biologic of rat fed four weeks with increasing amount of OTA were analysed. In families having forebears suffering BEN, OTA contamination occurrence was higher compared to non BEN families (3, 4). C8dG OTA adduct was detected in rat kidney as well as in human samples. OTB, OP-OA, 4-OH OTA, 10 OH OTA, two OTHQ metabolites (quinone form), DC OTHQ and GSH-OTA derivatives were identified in blood and urine. Female rat and women excreted faster and more efficiently OTA and OTA metabolites, whereas male rat and man accumulated OTA derivatives in blood. Simultaneous presence of other mycotoxins increases OTA-DNA adducts. Thus determination of OTA-DNA adducts and metabolites are relevant biomarkers for BEN.
