Sažetak
Yeast Sccharomyces cerevisiae is an unicellular model organism which is widely used for elegant research of fundamental processes in eukaryotic cells. Here we present some of our insights regarding gene targeting, chromosome duplication and essental genes in yeast. Gene targeting is the hallmark of gene therapy development. It is important to thoroughly understand the possible side effects and unwanted genetic events that can happen during gene targeting. We identified these aberrant events, of which we found the duplication of the entire targeted chromosome to be particularly interesting, because of the evolutionary importance of duplication events. In another research using the same model organism we investigated the fate of cells missing an essential gene. For this purpose 1095 strains, each missing a particular essential gene, were examined during sporulation. Very different dividing capacity and phenotypes of dying cells and colonies were observed. Generally, we concluded that "the most essenial" genes/proteins for germination of yeast spores are those involved in translation followed by protein synthesis, protein folding then transcription, while some metabolic processes are the least important.
