Sažetak
Aim was tTo test the efficacy of poly(hexamethylene biguanide) hydrochloride (PHMD) as a root canal irrigant against E.faecalis and P.aeruginosa in an experimental model ex vivo. Crowns and apical parts of twenty-two extracted single rooted human teeth were removed with diamond burs and root canals were instrumented with Hedström files to size #40. Smear layer was removed with sequential usage of 17% EDTA and 2.50% NaOCl and then sterilized in autoclave on 121ºC for 15 minutes. Specimens were randomly divided into even groups and mounted with sticky-wax at the bottom of cell culture wells. Eleven specimens were inoculated with an overnight broth culture of E.faecalis (ATCC 51299) and eleven with P.aeruginosa (ATCC27853). After 48 h incubation and irrigation with saline dentin samples were collected from inner root canal walls with Hedström files #50 before and after irrigation with 10 mL of 1% PHMD (2 mL every 2 minutes) followed by irrigation with 2 mL saline. Samples were put into vials containing 2 mL of sterile saline and tenfold dilutions, were plated on blood agar (Oxoid, UK) after 30s vortexing. Colony forming units (CFU) were counted after 48h incubation at 37ºC. Wilcoxon Signed Ranks test was used for statistical analysis. After treatment with PHMD P.aeruginosa was eradicated from all specimens and E.faecalis from 8 of 10 specimens. In two samples that grew E.faecalis after treatment reduction was from 104 CFU/mL to 101 CFU/mL. These results were statistically significant for both groups of microorganisms tested (P<0.01). PHMD in concentration 1% effectively reduces the number of E.faecalis and P.aeruginosa in an experimental tooth model and therefore its role as an endodontic irrigant should be studied further.
