Sažetak
Tumor associated glycoprotein 72 (TAG-72) is physiologically present in the secretory phase endometrium, but its presence and possible immunologic role in early normal human and tubal ectopic pregnancy (EP) decidua is largely unknown. The aim of the study was to analyze the presence of TAG-72 and its possible role on reproductive mucosal dendritic cells (DC) phenotype, cytokine expression and proliferation. The presence of TAG-72 at the implantation site was analyzed by double immunohistological labeling of TAG-72 and cytokeratin in normal and EP mucosal tissues. Flow cytometry was used for surface markers and intracellular cytokines detection in TAG-72 stimulated or un-stimulated DCs and for analysis of DC mediated T cell proliferation in the presence or absence of TAG-72. The double labeling of paraffin embedded early pregnancy decidua sections using B-72.4 anti-TAG-72 mAb and MNF 116 anti-cytokeratin mAb revealed the absence of TAG-72 in uterine decidua of normal and EP, although it was present in epithelial cells at and away from the tubal implantation site of EP. Decidual CD1a+ cells stimulated with TAG-72 decreased CD83 expression and diminished IL-15 and IFN- intracellular production. Mucosal DCs at the tubal implantation site express negligible IFN- and IL-15, whereas uterine DCs of EP show phenotype of normal pregnancy decidua. TAG-72 treated DCs increased cord blood T cell proliferation when compared to untreated DCs. TAG-72 did not change decidual T cell proliferation response. The anti-inflammatory properties of TAG-72- treated decidual CD1a+ cells in vitro is confirmed with characteristic of DCs at the tubal implantation site in the natural vicinity of TAG-72.
