Sažetak
Studies searching for human plasma N-glycans that change as a result of specific condition or disease were previously done on a small number of samples due to limitations of analytical methods. Recent improvement of analytical methods enabled our first large scale study on human plasma N-glycome. Results from almost 2000 individuals revealed high natural variability with magnitude that exceeds some previously described changes associated with diseases. The heritability of glycans was found to vary widely across individual glycan species. Effects of age, smoking, dietary habits and some other environmental factors were shown to have significant influence on specific glycan levels. On a smaller number of subjects, repeatedly sampled on a several occasions, stability of the N-glycome was determined in a single individual which leads to conclusion that N-glycome has a large biomarker potential. By involving multiple fluorophore labelling we have introduced a new approach enabling the increase of throughput from threefold on HPLC up to 36 fold on UPLC.
