Sažetak
Aldol addition is a reaction catalyzed by enzymes aldolases and cells use it for the formation of C- C bonds. In this work a novel reaction of aldol synthesis by aldol addition of dihydroxyacetone (DHA) to protected aldehide (N-Cbz-glicinal) catalyzed by enzyme D-fructose-6-phosphate aldolase (FSA A129S) [1] is presented. Addition reactions change the number of atomes or groups bounded to C-atomes. They happen on multiple bonds and are very frequent in metabolism (eg. Citryc acid cycle or carbohydrate methabolism). In this work D-fructose-6-phosphate aldolase derived from genetically modified E. coli (FSA A129S) was used which is functionally distinct from known aldolases in its tolerance of different donor substrates as well as acceptor substrates [2]. Kinetics of aldol adition of dihydroxyacetone to N-Cbz-glicinal was determined by initial reaction rates method. Measurements were conducted in batch reactor and the reaction was monitored by high performance liquid chromatography. From experimental data kinetic parameters of the model were estimated. Mathematical model was validated in batch reactor.
