Background and purpose: Naturally occurring cucumber mosaic Cucumovirus (CMV) and peanut stunt Cucumovirus (PSV) occasionally encapsidate satellite RNA (satRNA) along with genomic and subgenomic RNAs. CMV satRNA variants reported so far range in size from 330-405 nucleotides. PSV satRNA variants are, in average, longer (391-393 nt). To our knowledge there have been no reports of robinia mosaic Cucumovirus (RbMV) satRNAs. Materials and methods: A cucumovirus from Robinia pseudacacia was studied by the reaction of several ordinary test-plants, by means of biological separation of the virus from its satellite, and by double-stranded RNA analysis. Results: The type of symptoms in naturally infected Robinia and experimental hosts together with distinctive electrophoretic pattern leave no doubt about a cucumoviral, highly le likely RbMV, infection. The dsRNA patterns from originally infected Robinia leaf tissue or Nicotiana megalosiphon infected with original Robinia isolate showed also the existence of a satellite-like dsRNA band. It was slower compared to the CMV satellite dsRNA used as a marker. This satellite-like molecule was eliminated by serial inoculations through Chenopodium quinoa. Conclusions: The characteristics of investigated cucumovirus from symptomatic leaves of Robinia closely resemble RbMV. Its satellite-like RNA is longer than most CMV satellites and accordingly similar to PSV satRNA variants reported so far.
