Sažetak
Phytoplasmas are intracellular prokaryotic parasites/symbionts of the plant phloem and insect hemolymph. Grapevine yellows (GY) is economically important disease that affects grapevine (Vitis vinifera L.). Most important causative agents of GY in Europe are flavescence dorée (16SrV) and stolbur (bois noir, 16SrXII-A) phytoplasma. Molecular analysis of highly conserved 16S rDNA gene is not suitable to study detailed genetic variability and phylogenetic relationships among isolates from stolbur-group. Therefore, we have chosen a more variable non-ribosomal secY gene encoding a translocase protein to perform more detailed analyses among isolates of stolbur phytoplasma infected grapevine from Croatian vineyards. PCR products amplified using the PosecF3 and PosecR3 primers from the selected samples were cloned and sequenced. Obtained sequences were assembled and edited by Sequencher, while multiple sequences alignments were made using the ClustalX program. Phylogenetic analyses were conducted using MEGA 4. The obtained results showed considerable variability among isolates and proved that secY gene represents a valuable molecular marker for stolbur phytoplasma phylogenetic analysis.
