Sažetak
Citrus tristeza virus (CTV) is the causal agent of tristeza, the most devastating citrus viral disease. CTV may induce the development of three distinctive syndromes: quick decline, stem pitting and seedling yellows. Besides the rootstock/scion combination and environmental conditions, the major factor influencing the disease outcome is the viral genotype. CTV isolates can be classified in several phylogenetic groups based on the coat protein (CP) gene sequence. Since contradictory results are reported regarding the relationship between symptoms expression and isolate genotype, we analyzed symptoms in different indicator host plants caused by each molecularly characterized CTV monophyletic isolate (isolate with one virus genotype) separately. Based on the SSCP-analysis of these CP variants, they were separated by cloning, and sequenced. Six out of seven tested isolates clustered to the monophyletic groups Gp1, Gp2, Gp3a, Gp4, Gp5 and M and then were evaluated in graft-inoculated plants of Mexican lime, Madam Vinous sweet orange, sour orange, grapefruit and sweet orange plant budded on sour orange rootstock. Infected plants were observed for all changes that could be related to CTV-infection. The most severe reactions were associated with Gp1, Gp2, Gp3a and Gp4 isolates. Phylogenetic groups Gp5 and GpM displayed less severe pathogenicity reflected in the development of only one of the three syndromes mentioned above. In comparison with the pathogenic potential predicted from the literature, some of these isolates showed unexpected pathogenic effects.
