Sažetak
It is known that vegetative propagation of species in genus Betula by cuttings is of limited succes that are the main problem for foresters in mass propagation of selected trees. Yellow birch present not only economically important forest tree in North America but also ornamental species in Europe. Ever since, there are only a few reports consider this species in tissue culture. Therefore, the intention of our work was to investigate the regeneration ability of yellow birch. Dormant buds of 2-year-old seedlings of yellow birch (Betula lutea Michx.) were used as initial explants. The effects of growth regulators ; 1.3, 2.2, 4.4 mM BA, the combination of 2.2 mM BA with 2.9 mM GA3 and 0.1 mM TDZ and media ; mod. ACM, WPM and BTM, on shoot proliferation and elongation were tested. Mod. ACM medium supplemented with 1.0 mM IBA was used as a rooting medium. Shoot development in dormant bud culture was achieved on the complete ACM medium supplemented with 2.2 mM BA. On the mod. ACM, WPM and BTM media only 1-3 shoots with 3-5 nodal segments were developed. Shoot proliferation was not achieved when BA alone or in combination with GA3 was used. Adventitious shoot differentiation was stimulated when the mod. ACM medium contained TDZ. Rooting of shoots was successfully obtained on the mod. ACM medium supplemented with 1.0 mM IBA. The effect of the genotype on rooting was noticed. Plantlets were transferred into pots containing peat/perlite mixture, cultivated in a greenhouse and finally planted in the field. Our results showed that regeneration ability of yellow birch is lower in comparison to other birch species. Regarding to ability of adventitious bud regeneration with TDZ, we could suggest this approach for micropropagation of yellow birch.
