Sažetak
The RecBCD enzyme is involved in double strand break (DSB) repair in the bacterium Escherichia coli and posseses all the enzymatic activities for the formation of the RecA nucleofilament (helicase, single stranded endonuclease and RecA loading). The RecA nucleofilament is the signal for the SOS response. The SOS response involves the elevated expression of more than 50 genes with various functions that are induced in response to damage of chromosomal DNA. Genetic analyses have shown that RecA and RecBCD are essential for SOS induction in response to introduced DSBs. In some mutants which affect DNA repair and recombination, endogenous DNA damage can occur and those mutants display a constitutive SOS response (for example ruvA, recN, recG, dam etc). We wanted to test whether the RecBCD enzyme function is involved in producing the SOS constitutive signal. We measured constitutive SOS expression and SOS induction after introduction of a DSB in ruvA, recN and recG mutants. We found that both, constitutive and DSB mediated SOS induction are dependent on RecBCD function. Since they share the same genetic requirements, we propose that the major endogenous DNA damage responsible for constitutive SOS expression is related to DSBs. This result is in agreement with the opinion that collapsed replication forks are the major endogenous DNA damage since they have double strand DNA ends which are the natural substrates for RecBCD enzyme. This explans the requirement for the RecBCD enzyme in the constitutive SOS response.
