Sažetak
Quercetagetin-3-methyleter glucoside (3, 4', 5, 6, 7-pentahydroxi-3'-methoxyflavone-7-O-b-D-gluko-pyranoside) is the first flavonoid isolated from the Croatian endemic plant Centaurea rupestris L. In our previous investigations we established its antiviral, antibacterial and antifungal activities. In present study significant effect of quercetagetin-3'-methyleter and structurally related quercetin, on the intracellular calcium level of primary cortical rat neurons was established. The effect of quercetagetin-3'-methyleter was stronger in comparison with quercetin. Also, it was shown that the increase in effect was not proportional with applied concentrations of investigated flavonoids. Our results are in accordance with the results recently published by group from Finland regarding the effects of several flavonoids on the level of intracelular calcium in GH4C1 cells of rat hypophysis. The mechanism of action of isorhamnetin was also established by mentioned group. Based on the closely related chemical structure of isorhamnetin and quercetagetin-3'-methyleter the same mechanism of their action on the level of intracellular calcium could be supposed. Therefore, it is reasonable to assume that quercetagetin-3'-methylether, like isorhamnetin, induces increase in cellular cAMP which, through the action of protein kinase A, causes the opening of voltage regulated calcium channels.
