Sažetak
The aim was to determine the optimal conditions for braking the cell walls of fresh baker's yeast (Saccharomyces cerevisiae) ultrasonic homogenization and optimization process by response surface methodology in order to release intracellular contents and that wehen it comes to protein denaturation. The influence of process parameters, cycle 0.4-0.7-1 with amplitude 40-70-100%, the ratio S/L from 0.3-0.8-1.3 g + 20 mL of buffer and time consuming process of breaking up cell walls 50-10-15 minutes, in relation to the total concentration of protein. RSM to approximate the conditions for protein extraction (Cycle 1, the amplitude of 100%, the ratio of S/L 0.03 (0, 6 g +20 mL buffer) and time of 15 minutes) at which the model provided a maximum yield of protein 17, 38 mg/gs.t.. To describe the extraction of total protein was successfully used linear model showed good agreement with experimental data, in addition to not reached the maximum function, means are needed further experimental research to achieve the optimal conditions for breaking the cell walls of yeast.
