Sažetak
Metabolic Syndrome (MS) is recognized as a cluster of cardiovascular risk factors. All components of MS have a genetic base, among them are genes of the renin angiotensin system (RAS). We investigated whether gene polymorphism of angiotensin converting enzyme (ACE) and angiotensin II type I receptor (AT1R) increases susceptibility to MS. 225 individuals were studied including 120 patients with MS and 105 subjects without MS criteria. Methods: AT1R A1166C was genotyped by PCR-RFLP, and ACE I/D by PCR based methods. Results: Patients with MS were significantly different from subjects without MS in relation to mean body mass index (BMI), waist circumference (WC), systolic blood pressure, diastolic blood pressure, glucose, total cholesterol (C), triglycerides, HDL-C, and LDL-C (P<0.01). Genotype distribution for AT1R A1166C were in MS group: AA, 52.1% ; AC, 39.6% ; CC, 8.3%, and AA, 37.8% ; AC, 43.4% ; CC, 18.8%, in controls. The 1166CC genotype was associated with significantly lower body mass index (p=0.04) and also lower waist circumference and waist-hip ratio (p=0.03) in men. Additionally, C allele was associated with lower systolic blood pressure, and fasting triglyceride in both genders though these were not statistically significant. Genotype distribution for ACE I/D polymorphism in MS: 26.7% had an II genotype, 46.1% had an ID genotype, and 27.2% had a DD genotype ; in control group: II 31.2%, ID 50.4% and DD 18.4 %. The differences in the mean BMI, glucose, total cholesterol, triglycerides, LDL-C, and HDL-C were significant in patients with the MS and ACE DD genotypes (P<0.03). CONCLUSIONS: The AT1R 1166 CC genotype appears to predispose to favourable anthropometric and metabolic traits, relative to cardiovascular risk. The ACE DD genotype increases susceptibility to MS.
