Sažetak
Dalmatian black pine (Pinus nigra Arnold subsp. dalmatica (Vis.) Franco) is a native and endemic tree distributed particularly in the coastal regions of Croatia. The Dalmatian black pine is evergreen conifer species belonging to the family Pinaceae. Most conifer trees are considered a source of structurally diverse bioactive compounds, and have provided important contributions to the discovery of pharmaceuticals agents and other biomedical applications. Pine oils are widely used as fragrances in cosmetics, flavouring additives for food and beverages, scenting agents in a variety of household products, and intermediates in the synthesis of perfume chemicals. The rise in antibiotic resistant microorganisms in recent years has led to an increasing search for new antibiotics. The prevention and treatment of these infectious diseases by applying products from plants, as potential and promising sources of antimicrobial agents, has increased during recent years. Among these products, volatile oils from Pinus species have been shown to possess antibacterial and antifungal activities and could serve as a source of antimicrobial agents. Also, the majority of the preceding studies carried out with commercial volatile oils seldom compared the antimicrobial activity of pine oils on the basis of their chemical composition. However, there are no available reports on biological activities of Dalmatian black pine volatile compounds. Therefore, the aim of the present work was to study in vitro antibacterial and antifungal activities of the volatile compounds isolated from the fresh needles of Pinus nigra ssp. dalmatica respectively as related to their chemical composition. The chemical composition of essential oil was determined using gas chromatography–mass spectrometry (GC–MS) and flame ionisation detection (GC–FID). The main compounds were -pinene, ß-pinene, germacrene D and ß-caryophyllene. Disc diffusion and broth microdilution assays were used for in vitro antimicrobial screening. The essential oil of Dalmatian black pine exhibited great potential of antibacterial activity against Gram-positive bacteria (MIC = 0.03 - 0.50%, v/v) and lesser activity against Gram-negative bacteria (MIC = 2.5 - 3.2%, v/v). The volatile compounds also inhibited the growth of all fungi tested including yeast.
