Sažetak
Two groups of C57Bl mice, aged 8-10 weeks (5-8 animals per group) were fed, over 21 days, diets enriched with olive oil (5% addition to standard pellet, w/w), containing predominantly oleic acid (C18:1n-9), or standard pellet diet. The animals were one-third partially hepatectomised (1/3 pHx) under aether anaesthesia, sacrified by cervical disslocation 6th, 12th, 24th, and 48th hours later, when the brains were carefully removed. Total lipids were extracted from tissue samples with a chloroform/methanol (2:1, v/v) mixture according to Folch et al. Phospholipids (PL) were separated and purified from brain's total lipid mixture using solid phase extraction (SPE) procedure on aminopropylsilica columns. Fatty acids (FAs) of polar lipids were analysed in the form of fatty acid methyl esters (FAMEs) by gas chromatography (GC). Significant increases in the amounts of arachidonic and docosahexaenoic acids were detected in olive oil fed groups, 12 and 48 hours after pHx.
