Sažetak
The cultivation of Legionella on artificial media is complex, and for their cultivation BCYE (Buffered Charcoal Warner-Yeast Extract) agar and AYE (ACES buffered Warner Yeast Extract) broth are used. The aim of this work was to examine the growth curves of L. pneumophila and L. longbeachae during cultivation in AYE liquid medium. The effect of L-cysteine and iron salts on the growth of these strains was examined. The growth of L. pneumophila and L. longbeachae in nutrient broth was monitored spectrophotometrically, and by the tenfold dilution method. Generation time was determined by two methods, direct and indirect method. Our results showed that after 6 hours of lag phase, there has been exponential growth with generation time of 4:30 hours, or nearly 5 hours for L. pneumophila. After 18 hour of incubation the maximum number of cells was 109 CFU/ml. L. longbeachae remained in the lag phase for 8 to 9 hours, after which there was exponential growth of bacteria up to a maximum value of 5x108 CFU/ml. Amino acid L-cysteine was essential for the growth of both legionela, while supplemental iron was not essential but supported the growth. Cultivation of both strains showed the production of brown pigment in the stationary phase, although the L. longbeachae observed stronger pigment production. In conclusion, both legionela species are able to grow in AYE media and L-cystein is essential for their growth.
