Sažetak
Since it is well known that biliary obstruction induces a significant immune dysfunction, which might provoke serious clinical complications, in this study the immuno and hepato-corrective abilities of PGM-Zn were tested in mice subjected to common bile duct ligation. The data were evaluated by analysis of: a) phenotypic profile and cell cycle of intrahepatic, thymic and splenic lymphocytes, b) MHC class II expression in the liver and phagocytic ability of peritoneal macrophages and c) the dynamics of protooncogen products expression in the liver injured by cholestasis. The data have shown that obstructive jaundice induces an immunosuppression, which partially may be linked with the presence of inhibitors in cholestatic plasma. Pre-treatment with PGM-Zn, however, increased the proportion of lymphatic cells in S phase of cell cycle in thymus and spleen, increased the proportion of CD4-CD8- and decreased CD4-CD8+ T cells, and enhanced hepatic MHC-class II expression and phagocytic activity of peritoneal macrophages. As hepato-modulating effect of PGM-Zn the changes in the dynamics of the expression of c-fos, H-ras, K-ras, Ki67 and p53 in the liver were seen. In vitro, PGM-Zn was also able to abrogate the inhibitory effect of cholestatic serum on PMA and anti-CD3+PMA induced blastogenesis of human mononuclear cells.
