Sažetak
A series of the new synthetyzed N-sulphonylurea derivatives of pyrimidine nucleo-bases demonstrated antiproliferative effects against different human cancer cell lines, especially colon cancer cell lines. In this study, the anticancer effect of the investigated substances was evaluated by MTT reductions measurements in cancer cell lines and by radioalbeled thymidine incorporation studies. The tested compounds were very potent in suppresing the growth of the cell lines different hystological types of tumor such as MiaPaca 2, HeLa 2, Hep, CaCo2, MCT 8, SW 620, HT-29. The treatment with molar concentration of the substances (10-4-10-7) caused growth inhibition by more than 50%. The results revealed that the newly developed N-sulphonylurea derivatives have simillar or greated anticancer efficacy in compariision with 5-fluoruracil, currently used as a front line agent in many important tumors, particulary at high concentrations. We tested also antiproliferative effects of N-sulphonylurea enhancemended with azidothymidine and observed that N-sulphonylurea derivatives with azidothymidine in human colon carcinoma cells were most cytotoxic and have greater antiproliferative efficacy. Taken together, our results support the hypothesis that that investigated substances have synergistic with azidothymidine and have most effects on investigated tumor cells.
