Sažetak
Galectin-3 and galectin-1 are lectins that recognise β-galactoside units as their physiological ligands. Thus they are involved in a number of molecular mechanisms, such as cellular signaling, cellular adhesion and the regulation of apoptosis. The role of cytosine methylation in the CpG islands of the promoter region of galectin-3 gene, LGALS3 has just recently come into researchers' focus. This epigenetic mechanism is an important mean of silencing the gene expression and it is suggested that it might have an effect on the regulation of galectin-3 expression. In order to test the possibility of epigenetic regulation of galectin-3 and galectin-1 expression, we have treated THP-1, Jurkat and JCAM cell lines with 5-azacytidine, a cytosine analogue which causes unspecific demethylation of cytosines in DNA. The expression of galectin-3 and galectin-1 on the protein and gene level was monitored using Western blotting and real-time PCR technique, respectively. Following the treatment, no expression of galectin-3 was detected in cells that normally do not express galectin-3, Jurkat and JCAM. However, an increase of galectin-3 and galectin-1 expression was detected in THP-1 cell line. Even thought it can not be said with certainty that epigenetic mechanisms are the ones responsible for the changes in expression after 5-azacytidine treatment, results propose this as a possible scenario. Other mechanisms must not be disregarded.
