Sažetak
In order to identify developmentally important genes, especially those involved in nervous system development, we applied the gene trap method. The ES cells were genetically changed by a nonhomologous construct containing a splice acceptor and the promoterless genes lacZ and neoR. In case of an insertion within an endogenous gene the resulting protein is a fusion between upstreem part of the trapped gene and the inserted genes. On the basis of lacZ activity in 11.5 day embryos we selected the mouse lines showing restricted staining pattern, from which we continue to analyse three here diecribed lines.
Two of this lines named kosenic i lobel are expressed from E10.5 days onward n the CNS and the heart. Kosenic additionally shows expression within the pancreas, while lobel is as well expresed within the testis. Both genes are expressed in the brain of the adult mouse. In these two lines the gene trap insertion is within the previously unknown genes. The third line contains the insertion within the nucleolar proliferation associated gene P120, one of the most important markers of the nucleolus of the cells of malignant tumors. We showed that P120 is expressed during embryo development in all structures, but not in all cells. The crosses between P120 heterozigous animals resulted with no living homozigous offspring.
