Sažetak
Focal application of penicillin on the rat cortex is one of the most popular methods to study simple partial seizures. In low doses penicillin selectively blocks GABA- mediated IPSPs, but in higher doses actions are less specific. Thc EEG changes have shown major focal spike activity that closely resembled human interictal spikes. The aim of this study was to investigate whether nifedipine. nicardipine or nimodipine could prevent penicillin-induced changes in EEG power spectra. Experiments were performed on adult Wistar rats. They were anesthetized and placed in the sterotaxic apparatus. Vehicle or various doses (1 ; 3 ; 10 ; 30) of calcium channel blockers had been applied 30 minute prior to focal pcnicillin application (2000 IU) on the left motoric cortex. In each rat four screw electrodes were placed over the frontal and parietal cortex. The EEG was recorded in basal conditions and 5, 10, 15, 30 and 60 minutes after penicillin injection. In order to obtain EEG samples for frequency analysis, the output from EEG machine was amplified and transmitted to an IBM-AT compatiblc microcomputor, via A/D converter. The system was calibrated using 500 mikroV. Two 7.5 s samples of artefact free EEG were analyzed in all measurements for each experimental animal. In order to obtain power. spectra betwecn 0-30 Hz. spectral averages were computed using Fast Fourier Transform. Individual EEG power of each frequency bin was normalized to average total power of baseline. EEG frequencies were collapsed into 1 Hz bins. Our results showed that nifedipine and nicardipine in doses of 10 mg/kg and 30 mg/kg prevented the inerease in EEG power spectra caused by focal penicillin injection, while nimodipine was less effective.
