Sažetak
The purpose of the present study was to investigate the influence of the calcium channel blocker MK-801, on the Na+, K+- ATPase activity and JNK/c-Jun phosphorylation in rat hippocampus and cortex after exposure to global cerebral ischemia. Enzyme activity and activation of JNK/c-Jun were determined at different time points after ischemia (5 min, 10 min, 30 min, 6 hr, 12 hr, 24 hr or 72 hr). The same parameters are also determined in rats pretreated with different doses of MK-801 (0.03 ; 0.1 ; 0.3 or 1.0 mg/kg) 30 min before exposure to ischemia. We found transient increase of Na+, K+- ATPase activity after 30 min, but profound and significant decrease after 6 hr- 72 hr. Activation of JNK/c Jun axes was detected after period of 5 min and 10 min such as 6 hr and 12 hr, but the most significant increase was determined in early stage i.e. after 5 min reperfusion period. Pretreatment with 1.0 mg/kg of MK-801 preserved Na+, K+- ATPase activity and reduced JNK/c Jun activation up to contol levels in all tested groups. In conclusion, MK-801 showed protective efiect on the Na+, K+- ATPase activity and induction of JNK/c-Jun axis.
