Sažetak
Regeneration of shoots in stem callus cultures of the common oak (Quercus robur L.) was investigated. Internodal segments from in vitro propagated shoots (on GD medium supplemented with 3% sucrose, 0.7% Difco Bacto-agar and 0.9 µM BA) were subcultured on GD medium with different concentrations of BA (0.1, 0.5, 1.0 and 10.0 µM). After four weeks in culture two types of callus regenerated depending on the medium composition: a friable, light brownish coloured (0.1, 10.0 µM BA), and acompact nodular and brown (0.5, 1.0 µM BA). Established callus cultures were attempted by subculturing on GD medium with 0.9 µM BA. Calli initiated on media with 0.1, 1.0 and 10.0 µM BA died, but tissues initiated on medium with 0.5 µM BA regenerated one to six shoots per callus. Regeneration was achieved in 68 to 97% cultures, and was genotype dependent. Shoots were morphologically normal and could be microcloned on medium with 0.9 µM BA.
