Sažetak
Micropropagation of wild cherry (Prunus avium L.) was investigated. Initial explants, issued from the shoot apex or axillary buds, were taken from one year old plants (4 genotypes), five-year-old trees (2 genotypes), suckers initiated on roots (3 genotypes) and mature trees (2 genotypes). Uninjured buds were successively sterilized with 1% chlorine preparation (Izosan G) and 1% mercuric chloride. Contamination percentage was low. Culture initiatio success depended on the explant origin. Usin a mod WPM (Woody Plant Medium, developed by McCown and Lloyd) without chlorine ions and with NaFeEDTA 60 mgl-1 supplemented with 2.2 µM BA, 2.5 µM IBA and 0.3 µM GA3 it was possiblr to induce a very good multiple shoot induction (multiplication phase) with the rate of shoot multiplication from 3.9 to 6.6 for 4-5 weeks. The effect of different cytokinines (KIN, BA and 2iP) and auxins (IAA, IBA and NAA) and their concentrations on shoot multiplication was investigated. The greatest number of shoot was obtained on medium with 23.2 µM KIN and 5.7 µM IAA. When BA was used instead of KIN, shoots were vigorous.The medium with NAA caused a very poor shoot multiplication, but callus induction occured in 100% of. cultures. Before rooting, shoots can be elongated on the same medium with a lower concentration of BA (0.4 µM) and higher concentration of GA3 (2.9, 5.8, 8.7, 11.6, 14.5 µM). Shoots wre also elongated on a multiplication medium if the time between two subcultures was prolonged from 6 to 7 weeks. Elongated shoots (1.5-4 cm) were rooted on the mod. WPM with 4.9 µM IBA. Rooting depended on genotype and explant and was from 11-90.5%. Roots were also induced if shoots were put in a sterile mixture of compost, peat and perlite (1:1:1) after a treatment with 246 µM IBA durin 4 hours. Rooted plantlings were successfully adapted in greenhouse conditions. Regeneration of shoots from callus was obtained on the multiplication medium from three genotypes. The obtained shoots did not differ morphologically from the initial genotype.
