Sažetak
Our previous study indicated that the prevalence of common human papillomavirus (HPV) types in Croatia decreased with age, while the prevalence of rare HPV types increased. The objective of this study was to determine all HPV genotypes among women older than 55 years. Hundred and thirty six archival specimens collected from 1995 to 2010 were tested for the presence of HPV DNA by polymerase chain reaction (PCR) with three sets of consensus primers subsequently followed by type-specific PCR for the most common low-risk (LR) types (HPV 6, 11) and the most common high-risk (HR) types (HPV 16, 18, 31, 33, 45, 52, 58). Undetermined HPV types (samples positive with consensus primers and negative with type-specific primers) were subsequently sequenced at the local core sequencing facility (ABI PRISM 310 Genetic Analyzer, Applied Biosystems). Correlation between HPV genotypes and cytological diagnosis was assessed. According to Croatian cytological classification "Zagreb 2002", there were 28.7% ASCUS (atypical squamous cells of undetermined significance), 36% LSIL/CIN1 (mild dysplasia), 24.3% HSIL/CIN2 (moderate dysplasia) and 11% HSIL/CIN3 (high dysplasia) in the study group. Out of 136 cervical specimens, 53.7% were HPV DNA positive, of which 29.4% undetermined. HPV 16 contributed to only 4.4% cases, while HPV 31 was the most abundant with a prevalence of 7.4%, followed by HPV types 33 (4.4%), 6/11 (3.7%), 52 (2.9%), 58 (2.2%) and 45 (1.5%). The high prevalence of HPV 31 and HPV 52 (12.1% each) was associated with HSIL/CIN2 and the high prevalence of HPV 33 (13.3%) was associated with HSIL/CIN3. The distribution of other HPV types will be presented.
