Sažetak
In this study we used the FBKC rabies vaccine with antigenic value of 2, 7 IU/dose for vaccination of risk group in age range 23-58 years.The subject were vaccinated according to the WHO- recommended pre-exposure schedule, on 0, 7 and 28 days. They were divided into two groups - one receive standard 1, 0 ml doses of the vaccine intramuscularly in the deltoid muscle of the upper arm, the other receive 2x0, 1 ml of the vaccine intraderemally in the volar side of both forearm. All reactions were local and mild in 6, 25%, and dissapeared within 24 hours. The immunogenicity of the FBKC vaccine was assessed by the rapid fluorescent focus inhibition test (RFFIT) performed in Institute Pasteur, Paris, France (Centre National de Reference pour la Rage). Blood samples for serological testing were taken from all vaccinees on days 0, 14, 30, 90 and 1 year since the beginning of treatment. The kinetics of specific antibody elicitation was similar to that observed after application of HDCS or PCEC (Purified chick embryo cell) rabies vaccine. In all tested persons a protective level of rabies antibodies was detectable on the 14th day since primovaccination. The geometric mean titre (GMT) in the i.m. group on day 14 was 1, 24 IU/ml and in the i.d. group 2, 58 IU/ml of serum. On day 30the GMTs were 5, 15 IU/ml and 5, 85 IU/ml for the i.m. and i.d. group, respectively. On day 90 the values were 2, 33 IU/ml for the i-.m. and 3, 02 IU/ml for the i.d. group. Even after one year a protective level of rabies antibody was detectable in the sera of vaccinees. The level for the i.m. group was 1, o4 IU/ml and for they i.d. group 1, 18 IU/ml. The study has proven the FBKC vaccine to have a low reactogenicity and evoke high levels of rabies antibodies in immunised persons. It is therefore an important candidate vaccine for pre-exposure protection of risk groups applied by i.m. or by i.d. route.
