Sažetak
We have identified S. cerevisiae peroxin Pex21p as a protein that specifically interacts with yeast seryl-tRNA synthetase (SerRS). SerRS is an essential enzyme required for serylation of tRNASer. In higher eukaryotes, aminoacyl-tRNA synthetases (aaRSs) are known to participate in synthetase complex formation in order to enhance processivity and specificity of aminoacylation and protein translation, consequently. In many organisms, detected complexes contain nine aaRSs, specific for aminoacids Glu, Pro, Ile, Leu, Met, Gln, Lys, Arg and Asp. Except syntetases, all complexes include three other unknown proteins of molecular masses 18, 38 and 43 kDa. In yeast, several synthetases interact both with other aaRSs and with nonsynthetase proteins.
In order to find out whether SerRS participates in formation of such complexes, we screend yeast cDNA library using two-hybrid system. We found six potentially interacting proteins, five of which represent same ORF, yeast Pex21, protein involved in peroxisome assembly (Purdue PE (1998) J. Cell Biol. 143:7, 1859-1869). We checked whether these proteins interact in vitro, and studied the significance of that interaction.
