A synthetic two-stage procedure prepares a multifunctional chemical reagent (Figure 5) that is used in a novel way (Figure 1) to isolate and detect in mixtures of biological molecules specific biological receptors, lectins and other adhesion macromolecules, which bind in a weak and/or transient way to soluble small molecule ligands such as oligosaccharides, neuropeptides hormones or gangliosides in complex biological mixtures. In the first stage a derivative of digoxin is synthesised and in the second stage a ligand and a photoreactive cross-linker is added to the digoxin. After recognition of the ligand by its specific binding agent, the complex is illuminated with UV light and a covalent linkage is formed between the photoactive grouping and the bound partner. The complex is visualized or isolated using an anti-digoxin antibody. The bound molecule is released from digoxin under mild acid conditions.
