U OTO KOM I KONTINENTALNOM STANI`TU R HRVATSKE SA}ETAK Za hematoloake pretrage jelena lopatara ( Dama dama L.), uzimani su uzorci krvi i to 9 jelena sa oto ja Brijuni i 8 jelena iz privatnog uzgoja pored Bo~jakovine. Odreeni su slijedei parametri: RBC, Hgb, HMT, MCV, MCH, MCHC, RDW, PLT, MPV, i diferencijalna krvna slika. Dobiveni rezultati pokazuju razlike u hematoloakim parametrima kako izmeu mu~jaka i ~enki tako i izmeu jelena koji ~ive u kontinentalnom podru ju i onih koji ~ive na otoku. Hematoloake vrijednosti mu~jaka s Brijuna u odnosu na ~enke razlikovale su se samo u viaim vrijednostima trombocita u korist mu~jaka, dok su se vee razlike pojavile u vrijednostima izmeu mu~jaka i ~enki kontinentalnog staniata i to na na in da su ~enke imale viae vrijednosti eritrocita, hemoglobina i hematokrita, a mu~jaci viae vrijednosti leukocita. Komparirajui meusobno obje skupine jelena (otok - kontinent) o ita je razlika u svim istra~enim parametrima izuzev brojnosti leukocita. Diferencijalna krvna slika pokazala nam je izrazitu eozinofiliju kod kontinentalne skupine. Klju ne rije i: lopatari, hematologija, diferencijalna krvna slika UVOD Trauma, infektivne kao i parazitarne bolesti, manifestiraju se klini kim ili subklini kim znacima pri emu hematoloaki parametri mogu biti vrlo korisna komplementarna metoda u njihovu dijagnosticiranju ( Zomborsky i sur.1997). Specifi nost jelena u hematoloakom smislu ine eritrociti koji pored klasi nog bikonkavnog oblika mogu imati i oblik srpa ili lista. Pojava srpastih stanica u krvi ovjeka povezana je sa abnormalnoau hemoglobina S (HbS) i uzrok je ozbiljne, katkad fatalne anemije. Srpaste stanice u mnogih pripadnika porodice Cervidae, povezane su sa polimorfnim formama hemoglobina (Kitchen i sur. 1968). Postotak srpastih stanica u sika jelena (Cervus nippon) in vitro raste sa poveanjem pH i zasienosti kisikom. Ovaj proces je reverzibilan ( Ogawa i sur. 1991). Primjenom nekih sredstava za sedaciju kao ato je na primjer xylazine primjeena je pojava ni~ih vrijednosti eritrocita, hemoglobina, trombocita, RDW-a i limfocita, u odnosu na ~ivotinje kojima je krv vaena bez sedacije (Cross i sur. 1988). U literaturi nalazimo hematoloake vrijednosti za razne vrste jelena koje se razlikuju u ovisnosti od vrste (Hawkey i sur. 1985), dobi (Kovac i sur. 1997), podneblja i godianjeg doba ( Zomborsky i sur. 1997). Budui da kod nas sli na istra~ivanja nisu provedena ovi rezultati mogu poslu~iti kao okvirni prikaz standardnih hematoloakih vrijednosti jelena lopatara u Hrvatskoj, za navedena staniata. MATERIJAL I METODE Hematoloake pretrage obavljene su na dvije skupine jelena lopatara (Dama dama L.). Prva oto na skupina, od 9 jedinki ujedna ene starosti oko 11 mjeseci pripadala je populaciji koja se nalazi na Brijunima, na povraini od 50 hektara lovno produktivnog prostora. Druga skupina od osam jelena, starosti 2 do 3 godine pripada skupini koja se nalazi u kontinentalnom podru ju. Ova skupina boravi u ograenom prostoru veli ine 1,5 hektar na privatnom posjedu kod Bo~jakovine. Oto nu skupinu jelena inila su aest mu~jaka i tri ~enke dok su kopnenu skupini inili etiri mu~jaka i etiri ~enke. Uzorci za analizu prikupljeni su od oto na skupine jelena prilikom njihova odstrela, a krv je uzimana direktno iz srca. Kod kontinentalne skupine krv je uzimana po prethodnoj aplikaciji anestetika "Zoletil 100" s djelatnom tvari tiletamin i zolezepam (Virbac). Krv je vaena iz vene jugularis. U oba slu aja koriatene su epruvete sa antikoagulansom K - EDTA. Na hematoloakom broja u SERONO - 9120 Baker System odreeni su broj eritrocita (E), koncentracija hemoglobina (Hb), hematokrit (HMT), prosje ni volumen eritrocita (MCV), prosje na koli ina hemoglobina u eritrocitu (MCH), prosje na koncentrcija hemoglobina u eritrocitima (MCHC), distribucija eritrocita prema veli ini (rdw), broj leukocita (L), broj trombocita (T) i prosje ni volumen trombocita (MPV). Diferencijalna krvna slika odreena je iz krvnog razmaza obojenog po Pappenheimu. Statisti ka obrada podataka izvraena je na programu Statgraf. Rezultati Pojedina ni rezultati hematoloakih pokazatelja za svaku ~ivotinju prikazani su u tablicama 1 i 2. Diferencijalna krvna slika prikazana je u tablicama 3 i 4. razlike izmeu pojedinih skupina i lokaliteta prikaztani su u tablicama 5, 6, 7, 8 i 9. Svaka od tablica ima statisti ki obraene pokazatelje. Za razliek koriaten je Student T test. Pri procijeni vrijednosti uzeli smo u obzir lokaciju (otok  kopno), porijeklo jelena, na in dobivanja uzorka i spol. U dostupnoj literaturi nismo uspijeli pronai koji bi ukazivali na djelovanje anestetika  Zoletil 100 na promijene u vrijednostima hematoloakih parametara. Kod oto nih jelena nije uo ena statisti ki zna ajna razlika vezana za spol, osim u broju trombocita (400,83 x 109 /L  mu~jaci i 563,00 x 109/L ~enke) koji se i dalje kreu u okviru standardnih vrijednosti za jelene (220-570 x 109/L Hawkey 1982). Ostali parametri bili su neato ni~i od onih koje navode neki autori (Kova  i sur. 1997.) su dobili vrijednosti E-10,95 X10 12 /L, L  3,98 x10 9/L, Hb  1031 g/L i HMT  48 % dok (Zomborski i sur. 1997) navodi puno ni~e vrijednosti za E.- 6.1 do 9.0 x 10 12 /L, L  4,3 do 7,2 x 10 9 /L, Hb 159  166 g/L, HMT 29  44 % MCV 47-51 fL, MCH 19  27 pg i MCHC 361  567 g/L tako da se naae vrijednosti i dalje nalaze u zadenim okvirima. Hematoloaki parametri kopnenih jelena pokazuju statisti ki odreene znakovitosti odreene za spol. }enke imaju viae vrijednosti E - 10,30 x 10 12 /L, Hg-142,25 g/L, HMT 42,82, % i MCV 43,3 fL u donosu na mu~jake ije su vrijednosti E- 8.80 x 10 12/L, Hb 122,75 g/L, HMT-36,77 % i MCV 41,17 fL. Vrijednosti se kreu unutar rezultata koje su objevili veina autora. pojavu viaih vrijednosti E, Hb, HMT kod ~enki zamjetili su i (Zomborski i sur.1997).pa tako onui imaju vrijednosti za mu~jake E- 9,00 x 10 !2 /L, Hg- 110 g/L i HMT  43% dok ~enke imaju slijedee vrijednosti E-10,15 x10 12 /L, Hg 138 g/L i HMT 51%. Tu pojavu kod lopatara starijih od dvije godine tuma e utjecajem godianjeg doba (proljee i spolnim promjenama Zomborski i sur. 1997). Ukupana skupina kopnenih jelena pokazala je znatno viae vrijednosti E, Hb, MCHC, RDW i T u odnosu na oto ne, a neato manje razlike u vrijednostima HMT, MCV i MVH (tablica 7). Kod oto ne skupine mo~e se uo itin pojava viaih vrijednosti MCV pri ni~im vrijednostima E (E-7,41 x 10 12 /L pri MCV  44,23 fL) u odnosu na kopnene jelene gdje je broj E (E- 9,56 x10 12 /L pri MCV 42,24 fL). Takve podatke navode i (Peinado i sur. 1999). Razlike u gora navedenim vrijednostima izmeu kopnene i oto ne skupine jelena mogu se protuma iti na inom na koji je uziman uzorak krvi , nadmorskom visinom i ishranom. Budui da su oto ni jeleni odstreljeni, neki od njih su vjerojatno po eli krvariti prijew uzimanja uzorka krvi pa odtuda ukupne niske vrijednosti dijela hematoloakih parametara oto nih jelena u odnosu na kopnene. Komparirajui tablice 8 i 9 mo~emo uo iti zna ajnije razlike izmeu ~enki, otok-kopno, nego izmeu mu~jaka otok-kopno. }enke su pokazale zna ajnu znakovitost P manje 0.01 u broju E, Hb i RDW, a manju znakovitost P manje 0.05 u vrijednostima MVHC, i MPV. Kod mu~jaka P manje 0.01 je kod vrijednosti MCHC, RDW, i T, a Pmanje 0.05 kod MCV. Za jedne i druge spolne skupine zna ajna je razlika u vrijednostrima RDW i to kod ~enki otok-kopno (27,47-23,52%)a kod mu~jaka otok  kopno (27,12-23,10%) ato nam govori o heterogenijoj populaciji E kod ~ivotinja na otoku u odnosu na one na kopnu. Broj L podudara se sa onima drugih autora (Kovac i sur. 1997 2,9 do 5,0 x 10 9/l). Ovako niske naae vrijednosti L, (otok  4,02 x10 9/L i kopno  4,21 x10 9/L) kao i drugih autora nisu znak leukopenije. Smatra se da je leukocitni odgovor u divljaih pre~iva a ni~i nogo u domaih kao ato su krave i ovce (Hawky 1985). Razlika u naaoj diferencijalnoj krvnoj slici i one koju navodi Kovac i sur.1997 segmentirani- 40,5%, nesegmentiranih 6,4 %, eozinofila 3,7 % , limfocita 49,1 % i monocita 0,7% o ituje se u poviaenim vrijednostima limfocita 79,6% Za oto nu skupinu i izrazitu eozinifiliju 17,3 % za kopnenu skupinu jelena.Limfocitoza je karakteristi na za jelene (Hawky 1982, Neimi 1993) ali ovako visoki br, limfocita mo~e biti znak neke kroni ne upale ili reaktivnog odgovora limfocita naizgled zdravih ~ivotinja (Nemi 1993). Eozinofilija kopnenih jelena pokazatelj je prisutstva crijevnih nametnika (Nemi 1993). First group of 9 deer originated from Brijuni archipelago and second group of 8 deer originated from private farm in continental habitat. Blood samples of both sexes were taken due to haematological indices ( E-erythrocytes, L- leukocytes, T- trombocytes, Hb  haemoglobin, differential leukocyte count, MCV, HMT, MCH, MCHC, RDW and MPV). Results shoved differences in haematological parameters between males and females as well as between continental and island population. Haematological parameters of island males in compare to island females differ just in higher amount of trombocytes in males. Significant differences occurred in haematological parameters of continental population in manner that females had higher amount of E, Hg and HMT in compare to males. Comparing those two groups of fallow deer population, significant differences of their haematological parameters become obvious in most of them, except in number of leukocytes. Island population had mean value of L 4.02 x 109 L and continental population 4.20 x 109 L. U OTO KOM I KONTINENTALNOM STANI`TU R HRVATSKE SA}ETAK Za hematoloake pretrage jelena lopatara ( Dama dama L.), uzimani su uzorci krvi i to 9 jelena sa oto ja Brijuni i 8 jelena iz privatnog uzgoja pored Bo~jakovine. Odreeni su slijedei parametri: RBC, Hgb, HMT, MCV, MCH, MCHC, RDW, PLT, MPV, i diferencijalna krvna slika. Dobiveni rezultati pokazuju razlike u hematoloakim parametrima kako izmeu mu~jaka i ~enki tako i izmeu jelena koji ~ive u kontinentalnom podru ju i onih koji ~ive na otoku. Hematoloake vrijednosti mu~jaka s Brijuna u odnosu na ~enke razlikovale su se samo u viaim vrijednostima trombocita u korist mu~jaka, dok su se vee razlike pojavile u vrijednostima izmeu mu~jaka i ~enki kontinentalnog staniata i to na na in da su ~enke imale viae vrijednosti eritrocita, hemoglobina i hematokrita, a mu~jaci viae vrijednosti leukocita. Komparirajui meusobno obje skupine jelena (otok - kontinent) o ita je razlika u svim istra~enim parametrima izuzev brojnosti leukocita. Diferencijalna krvna slika pokazala nam je izrazitu eozinofiliju kod kontinentalne skupine. Klju ne rije i: lopatari, hematologija, diferencijalna krvna slika UVOD Trauma, infektivne kao i parazitarne bolesti, manifestiraju se klini kim ili subklini kim znacima pri emu hematoloaki parametri mogu biti vrlo korisna komplementarna metoda u njihovu dijagnosticiranju ( Zomborsky i sur.1997). Specifi nost jelena u hematoloakom smislu ine eritrociti koji pored klasi nog bikonkavnog oblika mogu imati i oblik srpa ili lista. Pojava srpastih stanica u krvi ovjeka povezana je sa abnormalnoau hemoglobina S (HbS) i uzrok je ozbiljne, katkad fatalne anemije. Srpaste stanice u mnogih pripadnika porodice Cervidae, povezane su sa polimorfnim formama hemoglobina (Kitchen i sur. 1968). Postotak srpastih stanica u sika jelena (Cervus nippon) in vitro raste sa poveanjem pH i zasienosti kisikom. Ovaj proces je reverzibilan ( Ogawa i sur. 1991). Primjenom nekih sredstava za sedaciju kao ato je na primjer xylazine primjeena je pojava ni~ih vrijednosti eritrocita, hemoglobina, trombocita, RDW-a i limfocita, u odnosu na ~ivotinje kojima je krv vaena bez sedacije (Cross i sur. 1988). U literaturi nalazimo hematoloake vrijednosti za razne vrste jelena koje se razlikuju u ovisnosti od vrste (Hawkey i sur. 1985), dobi (Kovac i sur. 1997), podneblja i godianjeg doba ( Zomborsky i sur. 1997). Budui da kod nas sli na istra~ivanja nisu provedena ovi rezultati mogu poslu~iti kao okvirni prikaz standardnih hematoloakih vrijednosti jelena lopatara u Hrvatskoj, za navedena staniata. MATERIJAL I METODE Hematoloake pretrage obavljene su na dvije skupine jelena lopatara (Dama dama L.). Prva oto na skupina, od 9 jedinki ujedna ene starosti oko 11 mjeseci pripadala je populaciji koja se nalazi na Brijunima, na povraini od 50 hektara lovno produktivnog prostora. Druga skupina od osam jelena, starosti 2 do 3 godine pripada skupini koja se nalazi u kontinentalnom podru ju. Ova skupina boravi u ograenom prostoru veli ine 1,5 hektar na privatnom posjedu kod Bo~jakovine. Oto nu skupinu jelena inila su aest mu~jaka i tri ~enke dok su kopnenu skupini inili etiri mu~jaka i etiri ~enke. Uzorci za analizu prikupljeni su od oto na skupine jelena prilikom njihova odstrela, a krv je uzimana direktno iz srca. Kod kontinentalne skupine krv je uzimana po prethodnoj aplikaciji anestetika "Zoletil 100" s djelatnom tvari tiletamin i zolezepam (Virbac). Krv je vaena iz vene jugularis. U oba slu aja koriatene su epruvete sa antikoagulansom K - EDTA. Na hematoloakom broja u SERONO - 9120 Baker System odreeni su broj eritrocita (E), koncentracija hemoglobina (Hb), hematokrit (HMT), prosje ni volumen eritrocita (MCV), prosje na koli ina hemoglobina u eritrocitu (MCH), prosje na koncentrcija hemoglobina u eritrocitima (MCHC), distribucija eritrocita prema veli ini (rdw), broj leukocita (L), broj trombocita (T) i prosje ni volumen trombocita (MPV). Diferencijalna krvna slika odreena je iz krvnog razmaza obojenog po Pappenheimu. Statisti ka obrada podataka izvraena je na programu Statgraf. Rezultati Pojedina ni rezultati hematoloakih pokazatelja za svaku ~ivotinju prikazani su u tablicama 1 i 2. Diferencijalna krvna slika prikazana je u tablicama 3 i 4. razlike izmeu pojedinih skupina i lokaliteta prikaztani su u tablicama 5, 6, 7, 8 i 9. Svaka od tablica ima statisti ki obraene pokazatelje. Za razliek koriaten je Student T test. Pri procijeni vrijednosti uzeli smo u obzir lokaciju (otok  kopno), porijeklo jelena, na in dobivanja uzorka i spol. U dostupnoj literaturi nismo uspijeli pronai koji bi ukazivali na djelovanje anestetika  Zoletil 100 na promijene u vrijednostima hematoloakih parametara. Kod oto nih jelena nije uo ena statisti ki zna ajna razlika vezana za spol, osim u broju trombocita (400,83 x 109 /L  mu~jaci i 563,00 x 109/L ~enke) koji se i dalje kreu u okviru standardnih vrijednosti za jelene (220-570 x 109/L Hawkey 1982). Ostali parametri bili su neato ni~i od onih koje navode neki autori (Kova  i sur. 1997.) su dobili vrijednosti E-10,95 X10 12 /L, L  3,98 x10 9/L, Hb  1031 g/L i HMT  48 % dok (Zomborski i sur. 1997) navodi puno ni~e vrijednosti za E.- 6.1 do 9.0 x 10 12 /L, L  4,3 do 7,2 x 10 9 /L, Hb 159  166 g/L, HMT 29  44 % MCV 47-51 fL, MCH 19  27 pg i MCHC 361  567 g/L tako da se naae vrijednosti i dalje nalaze u zadenim okvirima. Hematoloaki parametri kopnenih jelena pokazuju statisti ki odreene znakovitosti odreene za spol. }enke imaju viae vrijednosti E - 10,30 x 10 12 /L, Hg-142,25 g/L, HMT 42,82, % i MCV 43,3 fL u donosu na mu~jake ije su vrijednosti E- 8.80 x 10 12/L, Hb 122,75 g/L, HMT-36,77 % i MCV 41,17 fL. Vrijednosti se kreu unutar rezultata koje su objevili veina autora. pojavu viaih vrijednosti E, Hb, HMT kod ~enki zamjetili su i (Zomborski i sur.1997).pa tako onui imaju vrijednosti za mu~jake E- 9,00 x 10 !2 /L, Hg- 110 g/L i HMT  43% dok ~enke imaju slijedee vrijednosti E-10,15 x10 12 /L, Hg 138 g/L i HMT 51%. Tu pojavu kod lopatara starijih od dvije godine tuma e utjecajem godianjeg doba (proljee i spolnim promjenama Zomborski i sur. 1997). Ukupana skupina kopnenih jelena pokazala je znatno viae vrijednosti E, Hb, MCHC, RDW i T u odnosu na oto ne, a neato manje razlike u vrijednostima HMT, MCV i MVH (tablica 7). Kod oto ne skupine mo~e se uo itin pojava viaih vrijednosti MCV pri ni~im vrijednostima E (E-7,41 x 10 12 /L pri MCV  44,23 fL) u odnosu na kopnene jelene gdje je broj E (E- 9,56 x10 12 /L pri MCV 42,24 fL). Takve podatke navode i (Peinado i sur. 1999). Razlike u gora navedenim vrijednostima izmeu kopnene i oto ne skupine jelena mogu se protuma iti na inom na koji je uziman uzorak krvi , nadmorskom visinom i ishranom. Budui da su oto ni jeleni odstreljeni, neki od njih su vjerojatno po eli krvariti prijew uzimanja uzorka krvi pa odtuda ukupne niske vrijednosti dijela hematoloakih parametara oto nih jelena u odnosu na kopnene. Komparirajui tablice 8 i 9 mo~emo uo iti zna ajnije razlike izmeu ~enki, otok-kopno, nego izmeu mu~jaka otok-kopno. }enke su pokazale zna ajnu znakovitost P manje 0.01 u broju E, Hb i RDW, a manju znakovitost P manje 0.05 u vrijednostima MVHC, i MPV. Kod mu~jaka P manje 0.01 je kod vrijednosti MCHC, RDW, i T, a Pmanje 0.05 kod MCV. Za jedne i druge spolne skupine zna ajna je razlika u vrijednostrima RDW i to kod ~enki otok-kopno (27,47-23,52%)a kod mu~jaka otok  kopno (27,12-23,10%) ato nam govori o heterogenijoj populaciji E kod ~ivotinja na otoku u odnosu na one na kopnu. Broj L podudara se sa onima drugih autora (Kovac i sur. 1997 2,9 do 5,0 x 10 9/l). Ovako niske naae vrijednosti L, (otok  4,02 x10 9/L i kopno  4,21 x10 9/L) kao i drugih autora nisu znak leukopenije. Smatra se da je leukocitni odgovor u divljaih pre~iva a ni~i nogo u domaih kao ato su krave i ovce (Hawky 1985). Razlika u naaoj diferencijalnoj krvnoj slici i one koju navodi Kovac i sur.1997 segmentirani- 40,5%, nesegmentiranih 6,4 %, eozinofila 3,7 % , limfocita 49,1 % i monocita 0,7% o ituje se u poviaenim vrijednostima limfocita 79,6% Za oto nu skupinu i izrazitu eozinifiliju 17,3 % za kopnenu skupinu jelena.Limfocitoza je karakteristi na za jelene (Hawky 1982, Neimi 1993) ali ovako visoki br, limfocita mo~e biti znak neke kroni ne upale ili reaktivnog odgovora limfocita naizgled zdravih ~ivotinja (Nemi 1993). Eozinofilija kopnenih jelena pokazatelj je prisutstva crijevnih nametnika (Nemi 1993). Results shoved differences in haematological parameters between males and females as well as between continental and island population. Haematological parameters of island males in compare to island females differ just in higher amount of trombocytes in males. Significant differences occurred in haematological parameters of continental population in manner that females had higher amount of E, Hg and HMT in compare to males. Comparing those two groups of fallow deer population, significant differences of their haematological parameters become obvious in most of them, except in number of leukocytes. Island population had mean value of L 4.02 x 109 L and continental population 4.20 x 109 L. First group of 9 deer originated from Brijuni arcales and females as well as between continental and island population. Haematological parameters of island males in compare to island females differ just in higher amount of trombocytes in males. Significant differences occurred in haematological parameters of continental population in manner that females had higher amount of E, Hg and HMT in compare to males. Comparing those two groups of fallow deer population, significant differences of their haematological parameters become obvious in most of them, except in number of leukocytes. Island population had mean value of L 4.02 x 109 L and continental population 4.20 x 109 L. Differential leukocyte count shoved obvious eozinophilia in continental group of fallow deer. 13 N$hipelago and second group of 8 deer originated from private farm in continental habitat.
