Sažetak
The drug discovery process is long and expensive. With the aim to shorten this process by producing and/or collecting high-quality drug-like compounds, which generate quality valuable data from High Throughput Screening, about two years ago we established new PLIVA's Compound Library. Our HTS collection consists of over 25.000 organic compounds, produced by us or acquired from different companies or even collected from various sources. The Library consists of many different structural classes. These are handcrafted compounds, produced by original synthetic techniques, often unique to PLIVA. These techniques have been developed by us, based on 80 years of experience in medicinal chemistry. The compounds in our collection are diverse ; most of them follow Lipinski Rule of Five, which describes the characteristics of chemical compounds with a stronger prospect of becoming potential drug candidates relative to other molecules. But, rules are not absolute and should not be used to immediately eliminate compounds from consideration. Best exception of these rules is Azithromycin, successful “blockbuster” and powerful PLIVA-s antibiotic. We are trying to achieve high quality and have sufficient amounts necessary to perform screening on selected targets. Our compounds undergo rigorous analysis before becoming recorded as PL compounds (unique PLIVA's code). The structures are typically confirmed by LC-MS and H1NMR. Compounds are stored in vials and microtitar plates, and we are producing our own mother-plates for targeted screening. Our PLIVA Compounds Library and a data produced from HTS are important part of the PLIVA R&D Database, developed and maintained by Research Information Center, where many UserClient applications are built as a support of R&D.
