This invention relates to the manufacture of semisynthetic penicillins of the general formula I in which an amino acid salt of the general formula II is reacted with an N-chlorocarbonyl amide of the general formula III to form a mixed anhydride of the general formula IV and then subjecting the mixed anhydride IV to aminolysis by means of 6-aminopenicillanic acid of the formula V.
