Sažetak
The purpose of this study was to evaluate in vitro the cytotoxic effect of root canal filling sealers, AH+ and AH26. The experiments were performed on Chinese hamster fibroblasts V79. The materials were mixed, crushed, and extracted in dimethyl sulphoxide. Such extracts were incubated at 37°C for 1 h, 24 h or 7 days. They were added to V79 cell samples. After incubation for 72 h , the total number of cells, as well as the percentage of viable cells, were determined. During this period in the control samples 4.6 x 104 cells were grown. The results showed that both, AH 26 and AH+ were cytotoxic, and that AH+ was more toxic than AH26. No cells survived 1-h treatment with 16.7 ľg/ml of AH+, while with the same concentration of AH26, 77% and 85% of cells survived respectively. For both materials the effect was strongly dependent on concentration of sealer in the extraction solution. The materials extracted for 1 h were toxic. After 1-h treatment with 5.5 ľg/ml of AH+, 65% of cells survived, 53% after 24 h and 62% after 7 days, while 77%, 66% and 87% of cells survived respectively after treatment with AH26.
