Sažetak
The sulfonamide antimicrobials are a class of synthetic compounds derived from sulfanilamide. Sulfonamides are prescribed against a wide variety of bacterial infections, especially in the case of a potential hypersensitivity to penicillins. Very often prescribed sulfonamide antibiotic, in human and veterinary use, is sulfadiazine. Once released in the environment, substances may undergo different reactions resulting in partial or complete transformation and/or degradation of the parent compound. Degradation products are often potentially more toxic for environment than active substances. For monitoring of sulfadiazine active substance in environment and biological samples many references exist. But, only few reports have been published on the determination of sulfadiazine degradation products in the environment. [1, 2] The aim of this work was to find optimal conditions for mass spectrometry identification and determination of sulfadiazine and their degradation products (sulfanilamide, sulfanilic acid, aniline, hydroquinone, quinine, 2-aminopyrimidine and 4- amino-3-hydroxybenzensulfonic acid). All experiments were performed using HPLC-MS Varian L 1200 chromatograph via an electrospray ionization interface (ESI) in positive ion (PI) mode. Investigated parameters were needle and capillary voltage on mass spectrometer. Mass spectrometric conditions were optimized for each analyte through automated tuning procedures implemented in the instrument software. Optimal conditions were determined measuring breakdown curves for the protonated molecular ion ([M+H]+) of target substances at different spray voltages (1000-5000 V). Optimal mass spectrometry conditions for all investigated substances were: spray voltage 3000 V and capillary voltage 50 V. HPLC-MS method was validated for linearity, repeatability, limit of detection and quantification.
