Sažetak
The Alu family of short, interspersed repeats is found in all primate genomes with about 500.000 members distributed within the human genome (Deininger 1989). Some of human specific Alu elements are not yet fixed in human populations because of their recent amplification. Once an Alu element is inserted at a specific location, it appears not to be the subject to loss or rearrangement (Deininger 1989) � and that property makes them useful in human genetic studies. The aim of this study was to analyse Alu insertion polymorphisms of 3 loci (TPA 25, ACE, FXIIIB) in the sample of 105 examinees from the subpopulations of 6 villages on the island of Hvar, Croatia. Because of various ethnohistorical, economic and geographic reasons, subpopulations of the island of Hvar have remained reproductively isolated and therefore suitable for anthropological studies (Rudan et al. 1990). The DNA was extracted from the whole blood samples. Investigated polymorphisms were analyzed using PCR methods. Alleles were analyzed by agarose gel electrophoresis. The analysis of genetic structure involved allele frequencies determination by a direct counting method, testing of Hardy-Weinberg equilibrium, determination of coefficient of gene diversity � GST and coefficient of genetic kinship � RST. Genetic distances (standard Nei's and modified Cavalli-Sforza's) were computed and genetic kinship was estimated based on the method of Haprending and Jenkins. All three investigated loci follow the Hardy-Weinberg equilibrium. The value of Nei� s coefficient of gene diversity � GST varies from 4.2% (FXIIIB) to 6.1% (TPA 25). 5.2% of the total gene diversity is due to diversity between populations. The value of RST coefficient estimated from the matrix of genetic kinship corresponds with the overall GST value. The lack of statistically significant correlation among the matrices of genetic distances among the investigated populations of the island of Hvar show that they cannot be differentiated solely based on the analysis of Alu insertion polymorphisms.
