Sažetak
The aim was to explore the activation status of early decidual CD14+ cells in terms of surface markers expression, cyto/chemokine production, endocytic capability and lipopolysaccharide (LPS) response. Methods of the study: Labeling of CD14 and CD209 antigens was performed by immunohistology in the first trimester normal human pregnancy deciduas. Early pregnancy decidual mononuclear cells (DMC) were obtained by collagenase digestion and subsequent gradient density centrifugation. Gradient density separation was sufficient to obtain peripheral blood leukocytes. Flow cytometry was used to analyze phenotype and intracellular cyto/chemokine expression in un-stimulated or LPS stimulated CD14+ cells from deciduas or peripheral blood samples of the same women. Chemokine secretion was measured by ELISA in the supernatant of decidual magnetically enriched CD14+ cells. Endocytic activity was determinated by FITC-Dextran uptake using Flow cytometry in CD14+ cells, un-treated or treated with increasing concentrations of LPS. Results: Decidual and peripheral blood CD14+ cells expressed negligible percentage of CD1a and CD83 markers what distinguishes them from early dendritic cell differentiation stages. Decidual CD14+ cell population showed higher percentage of chemokine CC receptor 5 (CCR5) and decoy receptors (CD206, CD209, CD163), but they do not express CD16 in comparison to peripheral blood CD14+ cells. The dominance of IL-4+ over IFN-+ cells and CCL22+ over CCL3+ cells in decidual CD14+ cells was found. The secretion of CCL17 and CXCL10 was balanced. Decidual CD14+ cells internalized FITC-dextran efficiently, but they were incapable to decrease endocytosis or increase IFN- production after 4 hrs LPS - stimulation. Conclusions: Decidual CD14+ cells represent autentic decidual population, which is probably settled in decidual tissue from the peripheral blood due to their pro-inflammatory CCR5 expression. This population seemed to be alternatively activated within decidual tissue due to decoy receptor and CD16 expression pattern, as well as unresponsiveness to LPS stimulation. Intracellular cyto/chemokine production and secretion by decidual CD14+ cells suggest their anti-inflammatory orientation.
